Pharmacological Properties

 

Pharmacodynamics

 

Tiamulin belongs to the pleuromutilin class of antibiotics and exhibits bactericidal activity against susceptible bacteria at high concentrations. It exerts its antibacterial effect by binding to the 50S ribosomal subunit, thereby inhibiting bacterial protein synthesis. Tiamulin demonstrates excellent antibacterial activity against Mycoplasma spp. and Brachyspira hyodysenteriae (the causative agent of swine dysentery). It also shows good activity against most Gram-positive bacteria, including Staphylococcus spp. and Streptococcus spp. (except Group D streptococci). While it has some efficacy against Actinobacillus pleuropneumoniae, its activity against most Gram-negative bacteria is weak.

When combined with Chlortetracycline at a ratio of 1:4, this product is effective in treating bacterial enteritis, bacterial pneumonia, and swine dysentery caused by Brachyspira spp. in pigs. It is particularly effective against pneumonia caused by Mycoplasma or mixed infections involving Bordetella bronchiseptica and Pasteurella multocida.

 

Pharmacokinetics

 

Tiamulin fumarate is well absorbed following oral administration in pigs. The bioavailability is approximately 85% after a single dose, with peak plasma concentrations reached within 2–4 hours. The drug is widely distributed throughout the body, achieving the highest concentrations in lung tissue. It is extensively metabolized in the body, producing over 20 metabolites, some of which retain antibacterial activity. Metabolites are primarily excreted via bile in the feces, with approximately 30% excreted in the urine.

 

Drug Interactions

 

Polyether Ionophores: Concomitant use with polyether antibiotics such as Monensin, Salinomycin, and Narasin interferes with the metabolism of these ionophores. This can lead to toxicity in chickens, characterized by growth retardation, ataxia, paralysis, and even death.

Ribosomal Binding Agents: Concurrent use with other antibiotics that bind to the bacterial 50S ribosomal subunit (e.g., Macrolides, Lincomycin) may result in reduced efficacy due to competition for the same binding site.

 

Indications

 

Pleuromutilin antibiotic. Primarily used for the prevention and treatment of Chronic Respiratory Disease (CRD) in chickens, Mycoplasma pneumonia and Actinobacillus pleuropneumoniae in pigs. It is also indicated for swine dysentery and porcine proliferative enteropathy caused by Brachyspira spp.

 

Dosage and Administration

 

Route of Administration: Administration in drinking water.

Swine: 0.1–0.13 g per 1 L of water. Administer continuously for 5 days.

Chickens: 0.28–0.56 g per 1 L of water. Administer continuously for 3 days.

 

Adverse Reactions

 

When used at recommended doses, skin erythema (redness) may occasionally occur in pigs. Overdose can cause transient salivation, vomiting, and central nervous system depression in pigs.

 

Precautions

 

Contraindications: Strictly prohibited for use in combination with polyether ionophores such as Monensin, Salinomycin, and Narasin.

User Safety: Users should avoid contact with eyes and skin.

Swine: 7 days

Chickens: 5 days
