DESCRIPTION

 

Ketoprofen Injection 15% is a sterile, clear, colorless to pale yellow solution.

Chemical Name:

α α -Methyl-3-benzoyl-benzeneacetic acid.

Class: Non-steroidal anti-inflammatory drug (NSAID) of the propionic acid class.

INDICATIONS

 

This product is indicated for the control of inflammation and pain associated with musculoskeletal disorders and acute tissue injury in cattle and horses.

Musculoskeletal Disorders: Treatment of lameness, arthritis, and synovitis.

Acute Trauma: Management of pain and swelling resulting from soft tissue injuries, fractures, or surgical procedures (e.g., dehorning, castration).

Systemic Inflammation: Adjunctive therapy for acute mastitis and metritis to reduce fever and inflammation.

Respiratory Disease: Relief of clinical signs associated with bovine respiratory disease (BRD).

 

PHARMACOLOGY

 

Mechanism of Action: Ketoprofen exerts its anti-inflammatory, analgesic, and antipyretic effects by inhibiting the cyclooxygenase (COX) enzyme system. This inhibition blocks the synthesis of prostaglandins, which are mediators of inflammation and pain. Additionally, Ketoprofen has been shown to inhibit lipoxygenase pathways and reduce bradykinin levels, providing a dual mechanism of action that may reduce the risk of gastric irritation compared to other NSAIDs.

Pharmacokinetics:

Absorption: Rapidly absorbed after intramuscular or intravenous administration.

Distribution: Highly bound to plasma proteins (>99%). It penetrates well into synovial fluid and inflamed tissues.

Metabolism: Metabolized in the liver via glucuronide conjugation.

Excretion: Primarily excreted in the urine. The elimination half-life in cattle is approximately 4–5 hours.

 

DOSAGE AND ADMINISTRATION

Route of Administration: Intravenous (IV) or Deep Intramuscular (IM) injection.

Target Animals: Cattle, Horses.

Recommended Dose:

Cattle: 3.0 mg Ketoprofen per kg of body weight (equivalent to 2 mL per 100 kg).

Horses: 2.2 mg/kg body weight.

Frequency: Administer once daily.

Duration: Treatment should not exceed 3 to 5 days.

Administration Note: For IV administration, inject slowly. For IM administration, divide large doses into multiple injection sites to minimize local irritation.

 

CONTRAINDICATIONS AND WARNINGS

 

Hypersensitivity: Do not use in animals with a known hypersensitivity to Ketoprofen, Aspirin, or other NSAIDs. Cross-sensitivity may occur.

Gastrointestinal Ulceration: Do not use in animals with active gastric or duodenal ulcers or evidence of gastrointestinal bleeding.

Renal/Hepatic Impairment: Use with caution in animals with renal or hepatic dysfunction. Ensure animals are adequately hydrated to prevent renal toxicity.

Pregnancy: Use with caution in pregnant animals. Safety has not been established in pregnant cattle or horses.

Drug Interactions:

Do not use concurrently with other NSAIDs (e.g., Flunixin, Phenylbutazone) or corticosteroids, as this increases the risk of gastrointestinal ulceration.

May interact with highly protein-bound drugs (e.g., anticoagulants).

 

ADVERSE REACTIONS

 

Gastrointestinal: Anorexia, diarrhea, or gastrointestinal ulceration/bleeding (rare with short-term use).

Injection Site: Transient pain or swelling at the injection site, particularly with IM administration.

Systemic: Rare cases of anaphylaxis or renal papillary necrosis (usually associated with dehydration or overdose).

 

WITHDRAWAL PERIODS

Cattle (Meat): Do not slaughter for human consumption within 7 to 14 days of the last treatment (varies by region).

Milk: Do not use milk for human consumption during treatment and for 24 to 48 hours after the last treatment.

Horses: Not for use in horses intended for human consumption.

 

STORAGE

 

Store below 25°C (77°F).

Protect from light.

Keep container tightly closed.

Shelf life: Typically 24 months.
